Tobey Spider-Man took its cue from the Ultimate Marvel comic universe, as did the more recent Andrew Garfield flick, by trying to establish the nemeses between Spider-Man and Green Goblin/Norman Osborn by means of the Spider coming from Osborn's company, Oscorp. I didn't want to re-tread that old chestnut, so I figured Morbius made for a much better source of the genetic engineering that mutates the spider which in turn bites Peter. Especially since it was a similar accident that later makes Morbius into a vampire. 

However, in Stan Lee and Steve Ditko's original story, if memory serves, Norman himself was actually introduced after the Green Goblin, and Peter's line about how a man could hold off an army in the warehouse was lifted direct from the comic itself. 

As far as to why Macendale is the trigger man . . . well, as a certain Mazoku is fond of saying; that would be telling . . .
