The world being a rather large place probably has several "clans" of Devil
Hunters. Doesn't Yokho actually meet some of the "hunters" of the
mulltiveres in the second last or last ep?
Never having seen La Blue Girl (and planning to continue that trend), I can't
comment on the plot device question ... n.n;; But yes, Madoka says (or
implies) to Yohko that the Mano family is it. My first response to Mike's
question is that there are a couple of Mamono Hunters outside the Mano
family's main line (Kanzaki Azusa is in training; Mano Ayako was trying to
claim the title from Yohko but the two called a truce).
As for the "other Mamono Hunters" in OAV 5 ... eh, that's kind of
complicated. Also a spoiler. n.n;; The short version is that Yohko has met
Mano Haruka, the first Mamono Hunter; the others made a cameo appearance -
and they all lived in one timeline, one after the other.

Yohko *did* meet Mano Haruka, the first Mamono Hunter - she was trapped in an
alternate dimension of sorts, where she was battling Tokima; after Yohko was
killed and sent into the parallel dimension, Haruka brought her back (to
life, and then to their present-day Earth) and the two of them teamed up on
Tokima.
The other 105 generations of Mamono Hunters, or at least a great number of
them, were called up out of their times during that battle: Haruka used her
ring to summon them in response to Tokima's creating a horde of duplicates of
himself, and each Mamono Hunter re-inflicted the wound she'd given Tokima on
a corresponding Tokima-duplicate before vanishing (along with said
duplicate). While this was happening, Tokima himself was suffering the wounds
he'd collected over the centuries (again); finally, it fell to Yohko to
deliver the fatal blow and seal him away for good.
