I'm a fan of Jo Jo's, so I'll try and help answer your questions.  
(I'm also sending this to the list because I want to prove I haven't 
fallen off the face of the Earth and it might help someone else who 
wants to write a Jo Jo's fic. ^_^)

<1) How does one acquire a stand? Are they born with the ability, is 
it transfered to them somehow, or something else?>

They're born with it and it begins to manifest itself during the 
onset of puberty.

<2) What are the stands made of? Some, like Iggy's, were composed of 
their respective element, but what about one's like Jo-Jo's, Dio's, 
Polenareff's. ect?>

All Stands, even Iggy's, are psychic projections.  The Stands "forms" 
are mental manifestations of their powers, which can only be seen by 
other Stand Users.  Iggy's Stand is an exception because it uses an 
element, like the Water Stand user in the first two eps.
