It's said during the first story arc with the Dark Kingdom where they are
battleing Berryl.  During that time, Sailor V, AKA Sailor Venus was
supposed to be a decoy, something she took seriously as she was the one of
only four beings, including the crystal computer that had stored Queen
Selenity's spirit, to know who Usagi really was.  Usagi was said to be the
leader of the Senshi initially so that she could be protected from danger
while training her in how to be a good queen.  When the confrontation,
where Mamoru was harmed, happened, everyone regained their memories of
most of what had happened in the Silver Millenium.

Part of what had occured was the drastic change in Sailor V's and Sailor
Moon's attire.

Remember that Sailor Moon's costume was initially a virtual copy of Sailor
V's, right down to the mask that she wore.  A Change in the tiarra, where
the crescent moon appeared happened at their first meeting.  Soon after,
they were all taken to the Crown Games centre where they were sent to the
underground command centre.  It was then that Usagi decided that she would
be the best leader she could.

Because of this, she decided to send all the Senshi away, thus trying to
save their lives, and Venus protested, knowing the truth.  When it came
out, Luna was the one, who, thinking over everything to herself when Usagi
found Endymion gone, had cried herself to sleep, was watching her Princess
and thinking on what must be done.  She was the one who confirmed that
Sailor V, who's fortress survives on Venus, and who is the mortal
reincarnation of the Goddess Venus, is the leader of the Senshi.
