Definitely.  Minako is spelled with three characters - mi, na, and ko.
Minakou would be spelled with four - mi, na, ko, and u - which would
definitely imply both a pronunciation difference (you'd hold the 'o' sound
in Minakou longer than you would in Minako) and in the kanji (the 'ko' in
Aino Minako's name, which means 'child,' is never a lengthened 'o,' and as
such Minakou would need a different kanji in her name.)  Hope this helps!
